Limited sample volume can make mycoplasma testing especially difficult in cell therapy workflows, where material is often scarce and every milliliter matters. This application note presents a low-volume MycoSEQ Plus workflow evaluated in T cell containing matrices using 1–3 mL starting volumes. With manual or automated PrepSEQ extraction and ethanol precipitation, the method demonstrated detection of key mycoplasma species at 10 GC/mL, supporting low-volume testing strategies aligned with sensitivity expectations.
